Playters was the son of Sir Thomas Playters, 1st Baronet of Sotterley and his wife Anne Swan, daughter of Sir William Swan.<ref name=Burke>[https://books.google.com/books?id=K1kBAAAAQAAJ&dq=%22William+Playters%22+baronet&pg=PA415 John Burke, John Bernard Burke ''A genealogical and heraldic history of the extinct and dormant baronetcies'']</ref> He was knighted at Wanstead on 12 September 1623.<ref>[https://archive.org/stream/knightsofengland02shawuoft#page/n191/mode/2up Knights of England]</ref> He inherited the baronetcy on the death of his father in 1638.<ref name=Burke/>
In November 1640, Playters was elected Member of Parliament for Orford in the Long Parliament.<ref name=Willis>{{Cite Notitia Parliamentaria|converted=1|part=2|pages=229–239}}</ref> He was Deputy Lieutenant and Vice-Admiral of Suffolk between 1640 and 1649. He was also a colonel of a regiment until relieved of his posts by parliament.<ref name="suckling">Suckling. A. Playters married Frances Le Grys, daughter of Christopher Le Grys of Billingford, Norfolk. He had one son, Thomas, who died in 1651.<ref name=Burke/><ref name="suckling" /> He was succeeded by Sir Lionel Playters.<ref name=Burke/>
