A '''Spot-tag''' is a 12-amino acid peptide tag recognized by a single-domain antibody (''sdAb'', or nanobody). Due to the small size of a Spot-tag (12 amino acids) and the robust Spot-nanobody (14.7 kD) that specifically binds to Spot-tagged proteins, Spot-tag can be used for multiple capture and detection applications: Immunoprecipitation, affinity purification, immunofluorescence, and super-resolution microscopy.<ref name = "Virant_2018">{{cite journal |doi=10.1038/s41467-018-03191-2 |pmid=29500346 |title=A peptide tag-specific nanobody enables high-quality labeling for STORM imaging |year=2018 | last1=Virant |first1=David |last2=Traenkle |first2=Bjoern |last3=Maier |first3=Julia |last4=Kaiser |first4=Philipp D. |last5=Bodenhöfer |first5=Mona |last6=Schmees |first6=Christian |last7=Pisak-Lukáts |first7=Borbála |last8=Endesfelder |first8=Ulrike |last9=Rothbauer |first9=Ulrich | name-list-style = vanc |journal=Nature Communications|volume=9 |issue=1 |pages=930 |pmc=5834503 |bibcode=2018NatCo...9..930V }}</ref> Recombinant proteins can be engineered to express the Spot-tag.
