Micklethwait was educated at Shrewsbury School and at Magdalene College, Cambridge.<ref name="venn">{{cite web | url = https://archive.org/stream/p2alumnicantabri04univuoft#page/404/mode/2up | title = Alumni Cantabrigienses: Sotherton Micklethwait | author = J. Micklethwait graduated from Cambridge with a Bachelor of Arts degree in 1846.<ref name="venn"/> He was ordained as a Church of England clergyman and from 1849 to his death in 1889 he was vicar of Hickling in Norfolk.<ref name="venn"/> The benefice of Hickling was in the gift of the Micklethwait family and he was presented at his induction by his father, Nathaniel Micklethwait.<ref>{{cite news| title = The Church | newspaper = Cambridge Independent Press/British Newspaper Archive | page = 1 | location = Cambridge | date = 14 July 1849}}</ref>
