Singleton may have been the brother of William Singleton MP for Gloucester in 1640 and hence son of Thomas Singleton, merchant of London and Gloucester. He was Sheriff of Gloucester in 1634.<ref name=Williams>[https://archive.org/stream/cu31924030494953#page/n207/mode/2up William R Williams ''Parliamentary History of the County of Gloucester'']</ref> He was elected as an alderman in 1643<ref name="vch">{{cite book|title=VCH Gloucestershire Volume 4: The City of Gloucester|date=1988|page=377|url=https://www.british-history.ac.uk/vch/glos/vol4/pp374-381}}</ref> and served as mayor of Gloucester in 1645.<ref name=Williams/>
In 1659, Singleton was elected Member of Parliament for Gloucester in the Third Protectorate Parliament.<ref name=Willis>{{Cite Notitia Parliamentaria|converted=1|part=2|pages=229–239}}</ref> He was appointed a militia commissioner for Gloucester on 9 August 1659.<ref name=Williams/> He was removed from the city council in 1662.<ref name="vch" /> Singleton married Joan Robinson, daughter of Alderman Anthony Robinson.<ref name=Williams/>
