Mansel was the son of [Sir Lewis Mansel, 2nd Baronet](/source/Lewis_Mansel) of Margam and his third wife Lady Elizabeth Montagu, daughter of [Henry Montagu, 1st Earl of Manchester](/source/Henry_Montagu%2C_1st_Earl_of_Manchester). In 1660, Mansel was elected [Member of Parliament](/source/Member_of_Parliament) for [Glamorgan](/source/Glamorgan_(UK_Parliament_constituency)) in the [Convention Parliament](/source/Convention_Parliament_(1660)). He was re-elected MP for Glamorgan in 1670 for the [Cavalier Parliament](/source/Cavalier_Parliament) and sat until 1679. Mansel was an influential person in South Wales. The Duke of Beaufort, when [Lord President of Wales](/source/Lord_President_of_Wales), visited Margam during his "Progress" in 1684.<ref name=WBO>{{Cite DWB|id=s-MANS-OXW-1250|title=Mansel family}}</ref> Mansel died at the age of 69.

Mansel married Martha Carne, daughter of Edward Carne of Ewenny, Glamorgan. His son, [Thomas](/source/Thomas_Mansel%2C_1st_Baron_Mansel), succeeded to the baronetcy and was created [Baron Mansel](/source/Baron_Mansel). His daughter Martha married Thomas Morgan of Tredegar, which marriage and its after-effects caused the baronet much trouble because Thomas Morgan was reluctant to return from a tour on the continent of Europe.<ref name=WBO/> His daughter Elizabeth married [Sir Edward Stradling, 5th Baronet](/source/Sir_Edward_Stradling%2C_5th_Baronet).<ref name=Burke/>
