'''Dipeptidyl peptidase 8''' is an [enzyme](/source/enzyme) that in humans is encoded by the ''DPP8'' [gene](/source/gene).<ref name="pmid11012666">{{cite journal | vauthors = Abbott CA, Yu DM, Woollatt E, Sutherland GR, McCaughan GW, Gorrell MD |authorlink4=Grant Robert Sutherland | title = Cloning, expression and chromosomal localization of a novel human dipeptidyl peptidase (DPP) IV homolog, DPP8 | journal = Eur J Biochem | volume = 267 | issue = 20 | pages = 6140–50 |date=Nov 2000 | pmid = 11012666 | doi =10.1046/j.1432-1327.2000.01617.x | doi-access = free }}</ref><ref name="entrez">{{cite web | title = Entrez Gene: DPP8 dipeptidyl-peptidase 8| url = https://www.ncbi.nlm.nih.gov/gene?Db=gene&Cmd=ShowDetailView&TermToSearch=54878}}</ref>

This gene encodes a member of the [peptidase](/source/peptidase) S9B family, a small family of dipeptidyl peptidases that are able to cleave peptide substrates at a [prolyl](/source/prolyl) bond. The encoded protein shares similarity with [dipeptidyl peptidase-4](/source/dipeptidyl_peptidase-4) in that it is ubiquitously expressed, and [hydrolyzes](/source/hydrolyzes) the same substrates. These similarities suggest that, like dipeptidyl peptidase IV, this protein may play a role in [T-cell](/source/T-cell) activation and immune function. Alternatively spliced transcript variants encoding different [isoforms](/source/isoforms) have been described.<ref name="entrez" /> A relatively specific inhibitor of DPP8 and [DPP9](/source/DPP9), Val-boroPro <ref>{{cite web | title = PubChem:Val-boroPro| url = https://pubchem.ncbi.nlm.nih.gov/#query=Val-boroPro}}</ref>-,<ref name="pmid29396289">{{cite journal | vauthors = Okondo MC, Rao SD, Taabazuing CY, Chui AJ, Poplawski SE, Johnson DC, Bachovchin DA | title = Inhibition of Dpp8/9 Activates the Nlrp1b Inflammasome | journal = Cell Chemical Biology | volume = 25 | issue = 3 | pages = 262–267.e5 | date = March 2018 | pmid = 29396289 | pmc = 5856610 | doi = 10.1016/j.chembiol.2017.12.013 | url = }}</ref> leads to increased activation of the [inflammasome](/source/inflammasome) though both NLRBP1 and [CARD8](/source/CARD8) and can trigger [pyroptosis](/source/pyroptosis).<ref name="pmid29967349">{{cite journal | vauthors = Johnson DC, Taabazuing CY, Okondo MC, Chui AJ, Rao SD, Brown FC, Reed C, Peguero E, de Stanchina E, Kentsis A, Bachovchin DA | title = DPP8/DPP9 inhibitor-induced pyroptosis for treatment of acute myeloid leukemia | journal = Nature Medicine | volume = 24 | issue = 8 | pages = 1151–1156 | date = August 2018 | pmid = 29967349 | pmc = 6082709 | doi = 10.1038/s41591-018-0082-y | url = }}</ref>
