'''Claudin-5''' is a [protein](/source/protein) that in humans is encoded by the ''CLDN5'' [gene](/source/gene).<ref name="pmid9441748">{{cite journal | vauthors = Peacock RE, Keen TJ, Inglehearn CF | title = Analysis of a human gene homologous to rat ventral prostate.1 protein | journal = Genomics | volume = 46 | issue = 3 | pages = 443–9 | date = December 1997 | pmid = 9441748 | doi = 10.1006/geno.1997.5033 }}</ref><ref name="pmid9192844">{{cite journal | vauthors = Sirotkin H, Morrow B, Saint-Jore B, Puech A, Das Gupta R, Patanjali SR, Skoultchi A, Weissman SM, Kucherlapati R | title = Identification, characterization, and precise mapping of a human gene encoding a novel membrane-spanning protein from the 22q11 region deleted in velo-cardio-facial syndrome | journal = Genomics | volume = 42 | issue = 2 | pages = 245–51  | date = Sep 1997 | pmid = 9192844 | doi = 10.1006/geno.1997.4734 | doi-access = free }}</ref><ref name="entrez">{{cite web | title = Entrez Gene: CLDN5 claudin 5 (transmembrane protein deleted in velocardiofacial syndrome)| url = https://www.ncbi.nlm.nih.gov/gene?Db=gene&Cmd=ShowDetailView&TermToSearch=7122}}</ref> It belongs to the group of [claudin](/source/claudin)s.

== Function ==

This gene encodes a member of the claudin family. [Claudins](/source/Claudin) are integral membrane proteins and components of tight junction strands. Tight junction strands serve as a physical barrier to prevent solutes and water from passing freely through the paracellular space between epithelial or endothelial cell sheets. Mutations in this gene have been found in patients with [velocardiofacial syndrome](/source/DiGeorge_syndrome).<ref name="entrez" />

== Interactions ==

CLDN5 has been shown to [interact](/source/Protein-protein_interaction) with [CLDN1](/source/CLDN1)<ref name=pmid12909588>{{cite journal | vauthors = Coyne CB, Gambling TM, Boucher RC, Carson JL, Johnson LG | title = Role of claudin interactions in airway tight junctional permeability | journal = Am.
